Buying new tires for 2019 for my CX9
Taking off my 16,000 mile factory tires ... replacing with new ones. Just not me. Wondering if 275X45X20 will work with the standard drivetrain???? Anyone... thanks for your help!

You'll need to either find someone who has done this change, with a similar wheel size and tire size, or roll the dice and see what you get. Very possibly you'll be at risk for rubbing against either the fender at the wheel well and/or the suspension components inside.
Some references to consider.
Tire size comparison:
225/55R19 -- 4.9" sidewall; 28.7" diameter; 90.3" circumference; 702 revs/mile
275/45R20 -- 4.9" sidewall; 29.7" diameter; 93.4" circumference; 678 revs/mile; 3.5% over size
* Myself, I've often gone over or under by 1% or so, knowing full well it'll impact acceleration and the odometer/speed readings. But at much greater difference from stock, it's also likely to begin to affect handling in ways that can be detrimental. I couldn't say if you'll experience bad handling impacts with a 275/45R20 tire (with whatever rim you select). Beware of the potential risks.

On my own CX-5, I'd consider up to a 255 width tire (on a smaller-diameter rim than the factory spec). 235's currently. But whether it'd rub with a chosen rim (ie, 18x9 with appropriate offset) and a given tire is anyone's guess. Without finding a shop that's done this and some people who've done the tire I'm considering on that size of rim, I'd be on my own with respect to fitment. Might well end up with a set of tires that I can't mount and use. Such trial-and-error can rapidly get expensive.

Also... according to two tire size calculators.... pretty close I think... that's why I selected them... except they are almost an inch wider.
